Understanding HPLC Purity

High-Performance Liquid Chromatography (HPLC) is one of the most widely used analytical methods for assessing peptide purity. When a supplier lists a purity percentage (for example 98% or 99.9%), that number almost always comes from an HPLC analysis.

What HPLC Purity Actually Means

HPLC separates the components of a peptide sample based on their chemical properties. The main peak on the chromatogram represents the target peptide. The purity percentage is calculated by measuring the area of that main peak relative to the total area of all peaks detected.

Most peptide suppliers report purity using the formula:

Purity (%) = (Area of main peak ÷ Total area of all peaks) × 100

This is known as area-percent purity. It is the industry standard for research peptides, although it does not account for non-UV-absorbing impurities such as salts or residual water.
